Title: The glass nanopipette toolbox for biophysics and precision medicine

 

Abstract

The precision (personalized) medicine is a paradigm shift and provides exciting new opportunities for biomedical research. The rapid strides in nanoscience and nanotechnology have significantly contributed to the evolution of this field, giving rise to innovative approaches like DNA sequencing, wearable and portable sensors, and organ-on-chip devices. Originating from our endeavors in nanopore-based DNA sequencing, we have successfully developed a toolbox centering on multifunctional glass nanopipette, which is inexpensive, easy to use and widely accessible. By integrating nanoelectrode and nanopore at the nanopipette tip, the nanopipette can provide multiple modalities for precise molecule manipulation and sensing, enabling label-free, real-time and in situ biomolecule analysis and detection at the single-molecule level. Beyond DNA sequencing, we have applied the nanopipette techniques in diverse areas such as live cell/tissue imaging, quantitative intracellular delivery, intracellular single-molecule detection, and nanoelectrophysiology.
